Switchable tags

The following stocks carry transgenes with a cassette that can be inverted using FLP recombinase. As shown in this figure from Fendl et al. (2020) for the FlpTag transgene, the sequences for GFP will be spliced into mRNA when the cassette is inverted.

 FlpTag can be switched on or off by a recombinase.

The arrangement of the variant FRT sequences assure that the cassette will not reinvert.

If the FlpTag transgene is inserted in the opposite direction, GFP-tagged protein will be expressed until FLP-mediated inversion. In general, insertions that express tagged protein upon cassette inversion have “-X” in the insertion symbol.

The FlpTag.smV5 transgene contains a cassette encoding the V5 epitope in a nonfluorescent GFP backbone.

0, 1 and 2 denote reading frames, e.g. GFP is spliced into reading frame 1 when Mi{FlpTag.1} is inserted in an intron.
