Photoactivatable adenylyl and guanylyl cyclases

P{UAS-bPAC.S} encodes a bacterial adenylyl cyclase that converts adenosine triphosphate to cyclic adenosine monophosphate upon exposure to blue light. It was described in Stierl et al. (2011) “Light modulation of cellular cAMP by a small bacterial photoactivated adenylyl cyclase, bPAC, of the soil bacterium Beggiatoa” J. Biol. Chem. 286: 1181–1188.

and is present in the following stocks:


M{UAS-bPAC.M} is similar to P{UAS-bPAC.S}. M{UAS-bPGC.M} is identical to M{UAS-bPAC.M} except the adenylyl cyclase sequence was modified, as described in Ryu et al. (2010) “Natural and Engineered Photoactivated Nucleotidyl Cyclases for Optogenetic Applications” Journal of Biological Chemistry 285: 41501–4150, to produce an enzyme with high guanylyl cyclase activity and low adenylyl cyclase activity. Specifically, the enzyme has K197E, D265K and T267G amino acid substitutions.
